Overview

The KeyTec® TR-FRET AKT (S473) Phosphorylation Assay Kit is designed to measure the phosphorylation level of AKT at serine 473 (S473) in intracellular lysates. This kit employs a sandwich immunoassay format developed with KeyTec® TR-FRET technology, offering advantages such as simple and rapid operation, high specificity, high sensitivity, and excellent reproducibility. As illustrated in the schematic diagram, a pair of specific antibodies are conjugated to the fluorescence energy donor (Solar Eu1) and acceptor (LA2), respectively. Upon phosphorylation at the S473 site of AKT, both antibodies simultaneously bind to distinct epitopes on the AKT protein. Under external light excitation, energy resonance transfer occurs between the donor and acceptor. The phosphorylation level of AKT at S473 is then determined by measuring the signal intensity at the specific wavelength of 665 nm.
